Frequently Asked Questions

ELISA

Conjugate, substrate or wash buffer may be compromised. Washing may be excessive. Reagents may not have been brought up to room temperature prior to assay. Incorrect component or samples volumes may have been pipetted.

Washing may be inadequate. Incubation time and temperature may be too long or high. Incorrect component and samples volumes may have been pipetted.

Pipetting technique or automation instrument settings may need to be adjusted.

IFA

The microscope bulb maybe improperly aligned and/or the bulb maybe need to be replaced. Components may not have been stored according to package insert. Use humidity chamber for all incubation steps. Improper mounting medium may auto-fluoresce. Phosphate-buffered glycerol (pH 7.2) mounting media must be used.
Antigen may be compromised by condensation. Always brings slides up to room temperature before opening the slide pouch. Aggressive washing. Dirty glassware. Ensure that all glassware is clean and thoroughly rinsed. Excessive mounting media and/or poorly positioned cover slip.
Focus IFA controls come bottled at screening dilution. Do not dilute the IFA controls.

Molecular

The Integrated Cycler Studio Software has four fluorescence optic detection modules including, 520 nm (FAM or equivalent), 560 nm (JOE or equivalent), 610 nm (Cal Fluor Red or equivalent), 682 nm (Quasar 670 or equivalent).
No. The instrument is calibrated at the factory.
Yes. Assays can be defined and edited, and new runs setup while the instrument is in use. The assays will be saved until the instrument has completed its current run.
Yes. User-defined assay definitions can be exported and imported to instruments using the Manage Assays administrative functions.
Yes. The Thresholds can be adjusted while analyzing data for user-defined assay definitions.
PCR enzymes are heat labile. Store reagents as described in the product literature. Avoid freeze thaw cycles.
Decontaminate the work area, use fresh reagents, avoid cross-contaminating samples, and check sample order entered on Integrated Cycler Studio Software.
Contact the Lab Manager or Administrator.
The instrument has lost connection with the Integrated Cycler Studio Software. Verify connection or select FIND INSTRUMENT under the file menu.
Error messages are explained in the Integrated Cycler Studio Software Operator Manual.
